There is a line where Cletus is describing Mixmaster and he says it was designed specifically not to be able to merge human DNA with other species (probably as a safeguard to avoid lab accidents with the Proudfoot researchers).
He also mentions that any organism altered by Mixmaster will retain the compound in it's system/discarded cell tissue (like skin flakes etc) and when ingested by another organism, will absorb Mixmaster and become capable of merging it's DNA with another creature's DNA.
Which means, that by trying to eat 4-12, El Blanco should have absorbed Mixmaster into it's system. Even though El Blanco had to spit 4-12 out pretty fast thanks to 4-12's defense system, it was still long enough to have swallowed some of the defense secretions, which would have Mixmaster in them.
But apparently, El Blanco never mutated, so that would lead me to expect that Graboids are immune to the effects of Mixmaster.
Most likely because their DNA profile was unknown to the Proudfoot scientists and they couldn't make it able to have an effect on a creature who's DNA they didn't have samples of.

Mixmaster doesn't affect the organism that comes in contact with it, only its offspring, if I recall correctly. And El Blanco can not produce Shriekers since he is sterile.

Well, now that makes me wonder how Mixmaster can be used for proper genetic fusion.
If the compound doesn't mutate the organism it comes in contact with, but only changes the offspring, wouldn't that mean you'd have to dose up a normal animal or plant with it and then experiment on the offspring instead?
For example, to create 4-12 (who is mostly rhino), they'd have had to acquire an actual female rhino, use mixmaster on her and get a baby rhino from her with Mixmaster present in it's system?
It might have also required some invitro work, since Cletus mentions he carried 4-12 out in a satchel. And well, a normal rhino baby is a lot bigger then that!
Also, it appears there's a limit of around 3-4 DNA fusions that can occur before Mixmaster looses it's effect.
4-12 was roughly made from around 4 different kinds of animal.
The acid spitting plant was about 3 different kinds of DNA.
I'm assuming this is because absorbing and fusing too much DNA will create a non-viable lifeform, so Mixmaster can only do so much fusion before it becomes ineffective.
Also, it's possible that 4 fusions will deplete all the Mixmaster naturally present in the subject's body.

You can create something without needing a mother there. The creatures, including 412, were created in the lab. Basically, they were "test tube babies". The DNA from different creatures spliced together and then placed inside a surrogate mother, or some other sort of incubator.
And like Cletus said at one point, in the lab, they could control how mixmaster affected the creature; which they had to do otherwise they would get the crazy, unable-to-survive concoctions like those in the valley. So they could have easily controlled how big or small something would be.
